Neuroinflammation plays an important role in the pathogenesis of neurodegenerative diseases, including Alzheimer’s disease (AD) and ALS. Microglial activation, indicative of neuroinflammation, is present in patients with AD. One of Xona’s goals is to model critical aspects AD pathology in vitro to create simple yet robust models for therapeutics testing. Our methods rely on microfluidic compartmentalization to improve manipulations and access to cells to improve testing accuracy and throughput.

In a demonstration of our capabilities, we cultured human iPSC-derived eGFP-labeled cortical glutamatergic neurons (BrainXell) together with human iPSC-derived microglia (BrainXell) restricted to the axonal compartment using our XC450 XonaChips®. We activated microglia using lipopolysaccharide (LPS), a known inflammatory molecule. Using these human-derived cells, we recorded robust microglial activation (see figure). We found additional evidence of AD-like pathology in the somatodendritic compartment (not shown here).
